We find out in the episode 2.07 Darla that it was The Master who sired Darla, in a Virginia colony in 1609. Darla lay dying of syphilis at the time and was not afraid of the Master, who first enters her room dressed as a priest:
Darla: “My soul is well past saving. Let the devil take me if he’ll have me. Either way - I die.”
Priest: “No (The priest steps closer and pulls his hood back to reveal he’s the Master) you will not die. (Darla looks up at his inhuman face without flinching) You will be reborn.”
Darla: “I know you.”
Master: “I came to you last night. I sang to you from that window.”
Darla: “Hmm, I remember now. You’re death?”
Master: “No.”
Darla: “What then?”
Master: “I’m you’re saviour. God never did anything for you - but I will.”

Given that Jamestown was founded in 1607 and in 1609 was still struggling simply to remain in existence, I find it very unlikely that Darla, a prostitute, or the Master would’ve been there. The only it would have even been possible for a peasant prostitute, a dying peasant prostitute, to even get to Virginia would have been to go as an indentured servant, which would’ve been VERY unlikely. Her symptoms would’ve been far enough along to be felt, and quite possibly noticed by others, at least those who spent much time with her. That would’ve have made her less than desirable as a sexual servant, and one who simply wouldn’t live long enough to serve out her term anyway. Although it is possible her sponsor/owner wouldn’t have met her beforehand and therefore wouldn’t know about the disease.
The Master being there makes even less sense. Why travell from Europe with its millions people walking around like “Happy Meals with legs” (Spike)to go to a whole new continent with only two small settlements? He certainly wasn’t a Indian, and while it’s possible he travelled to America with the Spanish. Virginia is an awfull long way from any of the Spanish cities at the time. In all liklyhood the bulk of the Spaniards in America wouldn’t have even known where the English settlemants were, if they knew of them at all.
I just think that was a very poorly written part of the storyline. The writers apparently didn’t know thier history very well.

Also, how is Darla 400+ years old if she was sired in 1609? She would have been 388 when the show (Buffy)started.
